Application notes | The reactivity of this antibody with colon cancer extract and purified TAG-72 has been confirmed in solid-phase radioimmunoassays, and its reactivity with formalin-fixed carcinoma tissue sections confirmed in immunoperoxidase studies (Muraro et al, 1988). In flow cytometric analysis of freshly-isolated human carcinoma cells, this antibody reacts more strongly than the anti-TAG-72 MAb B72.3 (Muraro et al, 1988). Equally, flow cytometric analyses using this antibody have revealed that TAG-72 is expressed by certain haematopoietic cell lines (Nicolet et al, 1997). In western blot analysis, this antibody reacts with TAG-72 (Muraro et al, 1988). A humanised version of this antibody has been shown to be safe and sensitive in detecting recurrent colorectal carcinomas in radioimmunoguided surgery (Agnese et al, 2004), while radioimmunotherapy with an iodine-131-conjugated antibody has been shown to have a significant therapeutic effect on metastatic colon carcinoma in Phase I trials (Divgi et al, 1995). |
