Purity | Anti-Streptavidin antibody is an IgG fraction antibody purified from monospecific antiserum by a multi-step process which includes delipidation, salt fractionation and ion exchange chromatography followed by extensive dialysis against the buffer stated above. Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Rabbit Serum as well as purified and partially purified Streptavidin [Streptomyces avidinii]. No cross reactivity occurs against Avidin. |

Storage | Store vial at -20° C or below prior to opening. This vial contains a relatively low volume of reagent (25 µL). To minimize loss of volume dilute 1:10 by adding 225 µL of the buffer stated above directly to the vial. Recap, mix thoroughly and briefly centrifuge to collect the volume at the bottom of the vial. Use this intermediate dilution when calculating final dilutions as recommended below. Store the vial at -20°C or below after dilution. Avoid cycles of freezing and thawing. |

Biorbyt Rabbit anti Streptavidin and Biotin conjugated Rabbit anti-trypsin inhibitor antibody were used to detect target proteins Trypsin Inhibitor (left) and Streptavidin (right) under reducing (R) and non-reducing (NR) conditions. Reduced samples of purified target proteins contained 4% BME and were boiled for 5 minutes. Samples of ~1 ug of protein per lane were run by SDS-PAGE. Protein was transferred to nitrocellulose and probed with 1:1000 dilution of primary antibody (ON 4 C). Detection shown was using Dylight 649 conjugated Donkey anti rabbit (1:10K 1.5 hr RT in orb348637).

A three-fold serial dilution of DyLight™ 488 Conjugated Streptavidin starting at 200 ng was spotted onto 0.45 µM nitrocellulose and blocked in 1% BSA-TTBS (p/n orb348540, diluted to 1X) 30 min at 20°C. Anti-Streptavidin (RABBIT) Antibody (p/n orb344582) was incubated in Blocking Buffer for Fluorescent Western Blotting (p/n orb348637) at 1:1000 for 1 Hour at 20°C. An HRP Gt-a-Rb secondary antibody (p/n orb347654) was incubated at 1:40000 for 30 min at 20°C.

Expect: ~13 kDa reduced, ~60 kDa non-reduced.
