DescriptionInterleukin (IL)-15 is a 14-15 kDa four-helix bundle cytokine. IL-15, unlike other cytokines, is widely expressed by many cell types including monocytes, macrophages, dendritic cells (DCs), fibroblasts, epithelial cells, and skeletal muscle cells, but surprisingly not by T cells. It acts as a growth factor and also promotes the survival of T, B, and NK cells by preventing apoptosis through the upregulation of anti-apoptotic and downregulation of pro-apoptotic factors. IL-15 plays a crucial role in the development, homeostasis, and function of T, NK, and NK-T cells, and is additionally required for various functions of the B cells, DCs, macrophages, and mast cells. IL-15 is minimally secreted but is effectively delivered by trans-presentation in association with its unique receptor alpha (IL-15 Rα) on the surface of IL-15 producing cells to interact with a receptor complex composed of the IL-2R β and common γ chains on target cells. Trans-presented IL-15/IL-15 Rα signals through β and γ chains expressed on responding cells, leading to the recruitment and activation of JAK1 and JAK3. Activated JAK1 and JAK3 further phosphorylate STAT3 and STAT5, which prompts the transcription of IL-15-modulated genes in effector cells.
