Description | For proper targeting to the plasma membrane, the protein requires multiple cysteine residues. It is involved in the elimination of endogenous and exogenous organic anions in the kidney. This protein functions as an organic anion exchanger, where the uptake of one organic anion molecule is coupled with the efflux of one molecule of endogenous dicarboxylic acid (such as glutarate and ketoglutarate). It mediates the sodium-independent uptake of various compounds, including 2,3-dimercapto-1-propanesulfonic acid (DMPS), p-aminohippurate (PAH), ochratoxin (OTA), acyclovir (ACV), 3'-azido-3-'deoxythymidine (AZT), and others. The uptake of PAH is inhibited by several substances like p-chloromercuribenzenesulphonate (PCMBS), sulindac, and glutarate. Glycosylation at Asn-113 is important for proper targeting of this transporter to the plasma membrane. It belongs to the major facilitator superfamily and the organic cation transporter family. This protein is predominantly expressed in the kidney, with lower expression in the liver, skeletal muscle, brain, and placenta. It is localized at the basolateral membrane of the proximal tubule. |
